| Gene synthesis is the process
of template-free in vitro synthesis of a gene or other segment of double-stranded DNA. The most common methods involve oligonucleotide
synthesis and subsequent annealing of the resulting fragments, which are then ligated
and/or amplified by polymerase chain reaction (PCR).
Numerous companies worldwide provide commercial gene synthesis services. Gene synthesis is an important tool in many fields of recombinant DNA technology including gene optimization, heterologous gene expression, vaccine development, gene therapy, genetic engineering, synthetic biology, and synthetic genomics. De novo synthesis of custom optimized genes enables novel or improved functions or expression properties in functional genomics or synthetic biology applications. In many cases the synthesis of nucleic acid sequences is more economical than traditional cloning and mutagenesis procedures. |
